THE SUMMARY OF MY RESEARCH PLAN

Title          :  Pholymorphism Analysis of HSP 70 Gene in Brakel Kriel-Silver

Summary  : 
                      Brakel kriel-silver is chicken have double function is layer chicken and broiler chicken. Egg production of brakel kriel-silver about 190 - 250 grains per year. It is too good broiler. Male DOC breed for 2 - 3 months with good feed can 4 - 5 ons of body growth. But brakel kriel-silver easy to stress and it is seriously problem in brakel kriel-silver breeding of traditional breeding or conventional breeding. Because stress such as heat stress can causes decrease of production or productivity performance, live related with changing of physiological and biochemistry in it's body (Poultry Indonesia, 2011). Explained by Nataamijaya et.al (1990) on 25 - 31 dc of temperature shown decreasing eggs productivity and it's weight and slow growth. Side effect of heat stress is changing biochemistry such as decreasing of very low density lipoprotein (VLDL) and Vitellogenin, where it is used for egg yolk synthesis, so size of egg will be smaller than normal size. The method for increasing of production and productivity brakel kriel-silver is selection in moleculer level. Selection in this level especially HSP 70 gene for resistant of heat stress. The pholimorphism gene is very important for selection of brakel kriel-silver resistant of heat stress. But before selected, such as fenotype selection or moleculer selection must be analyzed of HSP 70 gene pholymorphism. So that very important to do research about HSP 70 gene variation with the title of research is "Pholymorphism Analysis of HSP 70 Gene In Brakel Kriel-Silver". This research aim to know mutation of HSP 70 gene in brakel kriel-silver chicken, where can used for refrence to analysis HSP 70 gene pholymorphism. The method used in this research is Single Strand Conformation Pholymorphism (SSCP) by virtue of Carmen Maria Mazzi, et.al (2003), where beginning by DNA extraction for separating of DNA and cell component. Furthermore Polymerase Chain Reaction (PCR) phase to do amplification of DNA target. Then Single Strand Conformation Polymorphism (SSCP) to viewing about HSP 70 gene pholymorphism by virtue of DNA migration. End of that is data analysis using Nei (1987) method.
