ANALISA PERBANDINGAN KENAMPAKAN DNA SAPI BALI DENGAN DNA AYAM ARAB PADA TAHAP VISUALISASI MENGGUANAKAN SINAR ULTRA VIOLET TERHADAP KEBERHASILAN PERCOBAAN PENELITIAN GEN HSP 70

ABSTRACT

In research conducted by scientists, research HSP 70 was not an easy thing. Because HSP 70 was a stranger among scientists, facilities used for research is very advanced and the issues that can not be resolved in this study. So before doing this research scientists have conducted experiments on penlitian of HSP 70.HSP research stage 70 includes several phases, namely stage of DNA extraction, PCR, and SSCP. These stages are very complicated stage in penelitian.Oleh therefore, very necessary dilakuakn experiment before doing research, especially in the extraction of DNA. In these experiments can be known how much ability in the use of tools, knowing the success rate of the method used and studied the errors in using the method. This study aims to determine the error on experimental methods of research that could cause any difference in appearance of the DNA and find out the cause of the difference in appearance of DNA in ultraviolet light. The results of this study indicate that in experimental studies on the addition of TE phenol phase retrieval errors of these materials. So that the stage of visualization using ultraviolet light, appearance of blood DNA Cows brighter than the appearance of blood DNA Arabic Chicken. Although the blood of cattle have been kept in a long time in the freezer. While the chicken blood DNA Arabs have a dirty appearance and looks like muddy. In addition, the number of visible bands of chicken blood DNA visualization Arabic only 1, while the blood of cattle blood DNA visualization there are two bands are visible in bright and clear. Also, look at some of the mistakes and errors in experimental research, there are several things that can cause failure in the visualization of DNA using Ultra Violet rays, including the addition of material error at the time of isolation, no precipitate DNA, DNA contamination, and problems with the pipette. Based on these results, it can be concluded that when visualized using Ultra Violet light, the appearance of bovine DNA more clearly and there are two bands are visible. While the chicken DNA Arab appearance looks a bit muddy and dirty and only seen one band Arab chicken DNA.
